How can I transfer my exporten local file to a aws Lightsail instance?

In order to make your website live on another hosting provider (outside of WP Engine or Flywheel which can be done with Local Connect) you should export your site and upload it to your host.
I would recommend using WP Migrate to export your Full Site. There are more details about how to do this here:
